A Cartilage Matrix Protein Regulates Collagen Synthesis in Mantle of Magallana gigas (Crassostrea gigas) under Ocean Acidification

The shell biosynthesis of oysters plays a critical role in protection against environmental stress, which cartilage matrix proteins (CMPs) determine the mineralogical and crystallographic properties shell. In present study, protein (designated as MgCMP1) was identified from Pacific oyster Magallana gigas (Crassostrea gigas) with objective understanding its possible formation. open reading frame (ORF) MgCMP1 1815 bp, encoding polypeptide 605 amino acids two von Willebrand factor (VWA) domains. mRNA transcript expressed constitutively all examined tissues higher level mantle, especially highest middle fold (MF) three folds mantle. addition, interaction between recombinant (rMgCMP1) bone morphogenesis 7 (rMgBMP7) vitro. After injection dsRNA to inhibit expression MgCMP1, Mgcollagen I X MF mantle significantly decreased. pre-puncturing acidification treatment (pH 7.8), thickness length new formation shells were lower than those control group 8.1), positive hybridization signals obviously weakened, MF, whereas decreased significantly. These results suggested that involved regulating response ocean (OA).
